From the 3gpp specification:
1 Radio Frame = 10 Sub-frame
1 Sub-frame = 2 Time-slots
1 Time-slot = 0.5 ms (i.e1 Sub-frame = 1 ms)
1 Time-slot = 7 Modulation Symbols (when normal CP length is used)
1 Modulation Symbols = 6 bits; if 64 QAM is used as modulation scheme
Radio resource is managed in LTE as resource grid..
1 Resource Block (RB) = 12 Sub-carriers

"Assume 20 MHz channel bandwidth (100 RBs), normal CP"
"Therefore,"
number of bits in a sub-frame = 100RBs x 12 sub-carriers x 2 slots x 7 modulatio
n symbols x 6 bits
= 100800 bits
"Hence, data rate = 100800 bits / 1 ms =100.8 Mbps"
"If 4x4 MIMO is used, then the peak data rate would be 4 x 100.8 Mbps = 403 Mbps
."
"If 3/4 coding is used to protect the data, we still get 0.75 x 403 Mbps = 302 M
bps as data rate."

this calculation is theoretical...if you need to calculate the actual throughput for LTE channel there are two methods ....

1.full simulation ( empirical )
2.simulation+mathematical formula (semi-empirical)

and that to consider all parameters that affect on LTE throughput which are:

I agree with aubidamax, LTE Throughput Calculation is much more complex. Simulations is best way.
But if you need robust calculation you can use Downlink Ring Method
(related doc: http://www.finetopix.com/showthread.php?28027-Search-latest-version-of-E*******-LTE-Coverage-amp-Capacity-Dimensioning),
The downlink ring method creates curves of throughput versus path loss, and
provides an alternative method of calculating cell throughput. The method
involves averaging the throughput over the cell area.

thanks for the above explantion..but could you please shed more light on DL throughput at PDCP, RRC, RLC and MAC layers with any examples? How would they differ and what needs to be considered for dl thput calculation at each layer?
