Hi!
Your question might be misunderstood.
In general, if you have 3D model of antenna radiation, you can calculate directivity by deviding signal strength in main lobe
with "average" signal strenght. (Average over all directions, thus estimating signal generated by isotropic antenna).
For dBi, you need to do 10*log_10 of Speak/Saverage.
Reasonable estimation can be given by calculationg just in two planes: H and V.
For TX antennas we can assume no loss, so Gain=Directivity.
In case you want to generate 3 D pattern from field measurments: you do not have enough samples to cover all directions,
but you can estimate by comparing with tuned radio prediction model.
